Título: ANTITUMOR EFFECT OF BETANIN-RICH EXTRACT OF OPUNTIA FICUS-INDICA FRUITS: THE MITOCHONDRIAL APOPTOTIC PATHWAY INVOLVEMENT

Resumo: Background: Opuntia ficus-indica (OFI) is rich in betanin, a water-soluble nitrogen-containing red-violet pigment, recognized for its antioxidant, cytoprotective, and anticancer properties. Objective: To evaluate the antitumor effect of betanin-rich extract (BRE) in cell lines and sarcoma 180 mice model. Methodology: The cytotoxicity of BRE, obtained via an aqueous two-phase system, was assessed in HCT-116, SNB19, and PC3 cell lines. Antitumoral effects were further investigated in thirty Swiss mice bearing transplanted sarcoma 180, randomly assigned into five groups: saline, 5-Fluorouracil (25 mg/Kg), and BRE (3, 10 and 30 mg/kg). The growth inhibition, tumours histopathological features, immunohistochemical expression of apoptotic markers, proliferation index, hematological and biochemical analysis were assessed after eight days. Results: In vitro, BRE exhibited over 75% inhibition of tumor cell growth. In comparison with the Saline group, BRE (30 mg/kg) significantly inhibited tumor growth in vivo (p<0.01) and increased the TUNEL, active caspase-3, and Bax immunoexpression (p<0.01). No significant differences were observed in Ki67 expression, haematological, biochemical, and histological parameters in the BRE-treated groups. Conclusion: BRE (30 mg/kg) possesses significant antitumor efficacy through activation of the mitochondrial apoptosis pathway. These results suggest BRE as a potential candidate for further exploration in chemotherapeutic and/or chemopreventive studies.
