Título: IMMUNOHISTOCHEMICAL EXPRESSION OF BRAF V600E MUTATION IN AMELOBLASTOMAS

Resumo: Objective: To investigate immunohistochemical expression of BRAF V600E mutation in ameloblastomas. Study Design: This is a retrospective cohort study. The study population consisted of individuals diagnosed with ameloblastoma. Clinical, imaging, histomorphological and immunohistochemical expression of BRAF V600E were analyzed. Bivariate analysis was performed through chi-square and Fisher's exact tests. Kaplan-Meier analysis with log-rank test and Cox proportional hazards regression were used to identify predictors of disease-free survival, with a statistical significance level of 5%. Results: Thirty-one individuals diagnosed with ameloblastoma were included. Tumors were more frequent in the 2nd and 3rd decades of life, with a male-to-female ratio of 1.5:1. BRAF V600E immunoexpression was present in 74,2% of the tumors, and was more frequent in men (p = 0.022). No association between BRAF V600E expression and other clinical, imaging and histomorphological variables was observed. Only the initial treatment modality was significantly associated with a better prognosis in univariate (p = 0.026) and multivariate (p = 0.021) analyses, with a Hazard Ratio of 22.14 (95%CI = 1.60-305.73) favoring radical treatment. Conclusion: BRAF V600E mutation is a frequent event in ameloblastomas. However, it does not seem to affect clinical behavior or prognosis in these tumors.
