Resumo: Oral cancer (OC) is the most frequent type of head and neck cancer, and it shows low survival rates when metastasis occurs. Tumor-derived exosomes induce a complex extracellular environment that could activate some proliferative and angiogenic pathways, starting a pre-metastatic niche. PI3k, Akt, and mTOR inhibitors have been studied as adjuvant therapy to improve antineoplastic treatment and reduce radiotherapy complications, such as radiation resistance and distant metastasis. This study aims to analyze the exosome expressions treated with PI3k, Akt, and mTOR inhibitors in the OC cell line. Exosomes were isolated from OC cell line (SCC-9) by ultracentrifugation, treated with PI3K inhibitor (LY249002), AKt inhibitor (Triciribine), or mTOR (AZD2014 - Vistusertib), and analyzed by Nanosight microscope. The PI3k inhibitor downregulated the liberation of exosomes, while Akt and mTOR inhibitors have no difference between DMEM/F12 medium and DMSO 0,1% and 0,5%. These findings demonstrate that PI3k inhibitor can reduce the exosomes from OC, and it is expected to provide an adjuvant therapeutic option against oral cancer proliferation.
