Resumo: Objective
We will evaluate the morphological, protein and metabolic changes presented in cell lines of COCE subjected to two models of cellular hypoxia.

Study Design
Prospective observational descriptive study
We will use the method of chemical hypoxia induced by cobalt chloride (CoCl2) and the coverslip variant model in different cell lines (HaCaT, Cal 27, SCC9 and SCC25). The study groups are: Control group (normoxia), CoCl2 Group, and coverslip variant Group. Tests will be performed after 24h of hypoxia. Morphological changes and dynamic behaviours will be evaluated by microscopic analysis. Furthermore, we will measure levels of hypoxia-associated protein (HIF-1α) by immunocytochemistry, metabolic changes of lipid droplets by Nile Red, and Nanolive 3D Cell Explorer microscope. Oxidative stress will be evaluated by ROS tracker dye.

Results
Both models induce changes in cell morphology in oral carcinoma cell lines by inducing hypoxia for 24 hours. Furthermore, it was observed the formation of lipid droplet in both situations. However, this effect was more evident in the coverslip method compared to CoCl2 method.

Conclusion
Our preliminary results suggest that coverslip hypoxia, and CoCl2 are suitable models to characterize the physiological changes to better understand how oral carcinoma cells respond to and adapt to hypoxia.
