Título: ANTITUMOR ACTIVITY OF HEXANE EXTRACT OF MIKANIA GLOMERATA LEAVES RICH IN ENT-KAURENOIC ACID COMPLEXED IN CYCLODEXTRIN

Resumo: Objective. The purpose of this study was to investigate the antitumor activity of inclusion complexes formed with hexane extract of Mikania glomerata leaves rich in ent-kaurenoic acid and β-cyclodextrin (ERKA/ß-CD) in rodents. Study Design. ERKA/ß-CD obtained by malaxing method was characterized regarding their physical properties, encapsulation efficiency, and cytotoxicity for L929 and MCF-7 cell lines using MTT assay. The antitumor activity was performed in mice with transplanted sarcoma 180 treated with saline, 5-fluouracil (5FU) and ERKA/ß-CD (30, 100 and 300 µg/kg, intraperitoneal) for seven days (ethical approval 030815). The inhibition growth, pathological features, and TUNEL, ki67, NFkB and NRF2 expression were assessed in the tumors. Serum lactate-dehydrogenase activity (LDH) was also evaluated. Results: Thermal analysis and FTIR confirmed the inclusion complexes formation, which showed strong cytotoxicity against MCF-7 (79.6 ± 3.1%), but weak against L929 cells (34.7 ± 4.9%; p<0.001). The encapsulation efficiency was 65.37 ± 4,72%. ERKA/ß-CD 300 significantly reduced tumor growth, mitotic activity and necrosis, increased TUNEL-detected cell death, and reduced Ki67 and NFkB immunoexpression, and serum LDH activity compared with saline (p<0,05). Conclusion. The ERKA/ß-CD inclusion complexes at 300 µg/kg displays antitumour activity in mice, likely due to inhibition on the NFkB signaling pathway and LDH activity.
