Título: SPATIAL LOCALIZATION OF MACROPHAGES AND GALR2/GALANIN EXPRESSION IN ORAL SQUAMOUS CELL CARCINOMA (OSCC)

Resumo: Objective: Investigate the spatial localization of macrophages and the expression of GALR2/Galanin in the tumor microenvironment of OSCC.
Study Design: Serial sections of surgical specimens of 18 OSCC cases (n=9 T1/T2 and n=9 T3/T4) were obtained and processed for multiplexed immunofluorescence analyses of CD68, CD206 and Cytokeratin by hybridization chain reaction. Conventional immunostaining was performed for Collagen IV and CD14 immunoreactivities. Prevalence and spatial distribution of CD68+/CD206+ macrophages were correlated with TNM clinical staging. RT-qPCR was performed using RNA extracted from the same specimens to assess the expression of GALR2 (considered an oncogene in HNSCC) and Galanin (GALR2 ligand).
Results: Collagen IV was stained to assess the integrity of the epithelial basement membrane allowing the identification of tumor invasion. CD206+ cells were closely associated with tumor cells (Cytokeratin+) in areas of invasion. Increased number of CD14+ cells were also positively correlated with tumor size and with less differentiated tumors. Increased expression of GALR2 and Galanin was associated with larger tumors.
Conclusion: CD206+ macrophages are associated with areas of tumor invasion. GALR2 expression and prevalence of CD14+ cells are greater in larger OSCC lesions.
