Resumo: Objective: To evaluate the immunohistochemistry expression of angiotensin-converting enzyme (ACE) 1 and 2 in oral squamous cell carcinomas (SCC). Study Design: Retrospective cross-sectional study based on clinicopathological and immunohistochemical analyses of 30 samples of oral SCC and 10 samples of healthy oral mucosa. The immunohistochemical evaluation was performed using a semi-quantitative scoring system, which analyzes the percentage of positive cells and their staining intensity to obtain the staining index (CI). Statistical analysis was performed to evaluate the associations between the clinicopathological and immunohistochemical variables (p < 0.05). Results: ACE 1 and 2 were expressed, in different percentages and intensities, in the cytoplasm of tumor and stromal cells, particularly fibroblasts present in the tumor microenvironment. ACE1 expression were higher in tumor cells (p<0.05) and ACE2 in healthy tissues (p<0.05). There was association between ACE1 expression and poorly differentiated tumors (p=0.001), and immunostaining of ACE1 and 2 in tumor cells was positively associated with immunostaining of these proteins in underlying stromal tissues (p <0.05). Conclusion: ACE1 expression was higher in poorly differentiated tumors. Further studies are needed to evaluate this marker as a potential prognostic marker, besides the possibility of using the renin-angiotensin system enzymes as probable therapeutic targets in oral SCC.
