Título: THE EFFECT OF EPIGENETIC DRUGS INHIBITION ON CANCER STEM CELLS IN ADENOID CYSTIC CARCINOMA LESIONS OF THE SALIVARY GLAND

Resumo: Objective: analyze the inhibitory effect of histone deacetylases on cancer stem cells from Adenoid Cystic Carcinoma (ACC) using a 3D high throughput tumor sphere technology. Study design: we used the UM-HACC-2A cell line to seed 5000 cells per well in a 96-well round-bottom plate assisted by an automatic liquid handler. Tumor cells were incubated for 3 days without disruption to allow the formation of spheroids. On day four, an epigenetic drug library was pipetted using an automated liquid handler, and on day six, Hoechst 33342 and Propidium Iodide were added to spheroids. All the spheroids were analyzed based on cell growth and cell death from 0 hours to 48 hours after the addition of drugs using a high throughput confocal imaging system. Results: the epigenetic modification library revealed about seventy-five UM-ACC-2A tumor spheroids present within the 25 top percentile of the cell death group. Among the target groups causing epigenetic modifications, 42.6% were HDAC inhibitors, followed by histone methyltransferase inhibitors (17.3%) and bromodomain inhibitors (7.5%). Conclusion: our results are promising because, for the first time, epigenetic drugs are being screened specifically for ACC tumors. Next, we will validate the top pharmacological candidates and test these drugs in patient-derived xenografts.
