Título: A PROPOSAL FOR AN EFFICIENT AND SECURE PROTOCOL FOR STUDYING ORAL CARCINOGENESIS WITH 4-NITROQUINOLINE-1-OXIDE (4NQO)

Resumo: Objective: The aim of this study was to propose an adapted effective and secure methodology for studying oral carcinogenesis throughout 4-nitroquinoline-1-oxide (4NQO). Study Design: Twenty-two male Wistar rats were exposed to 4NQO, evaluated clinically once a week, and euthanized at 12 and 20 weeks for histopathological analysis. The protocol involves a staggered dose of 4NQO up to a concentration of 25 ppm, associated with two days of pure water, a 5% glucose solution once a week and a hypercaloric diet. Results: At week 7, all animals displayed clinically evident tongue lesions. From a histological perspective, after 12 weeks of 4NQO exposure, 72.7% of the animals developed dysplasia and 27.3% developed in situ carcinoma, whereas in the group exposed for 20 weeks, dysplasia and in situ carcinoma were diagnosed in one case each, and carcinoma was diagnosed in 81.8%. No chronic weight loss or death were observed. This modified protocol ensures the development of oral lesions and prevents the adverse effects of the carcinogen. Conclusion: This new proposed 4NQO protocol was effective and secure for studying oral carcinogenesis and can be used to conduct lengthy investigations.
