Título: Oral squamous cell carcinoma cancer stem cells are strongly sensitive to NFκB inhibition

Resumo: Objective:Oral squamous cell carcinoma(OSCC) presents a modest response to available systemic treatments, which could be associated with cancer stem cells(CSC). We evaluated the role of NFκB and histone deacetylation(HDAC) inhibitors, administrated as a single agent, as a two-hit approach on the behavior of CSC derived from OSCC.Study design: SCC4, SCC9, SCC25 OSCC cell lines were used. CDDP(IC50) was determined by MTT assay. CSC presence was assessed by tumorsphere. Results:CDDP administration showed reduced viability of OSCC cells and increased CSC. A single and low administration of emetine and SAHA were able to disrupt CSC by inhibiting the NFκB pathway and increasing the histone acetylation levels, respectively. Further, when emetine and SAHA were administered together drives to the same CSC disruption seen after a single shot of emetine alone. Conclusion:Emetine showed superior anti-neoplastic properties in CSC OSCC cell lines than SAHA, by inhibition of NFκB, with effective inhibition of tumorspheres formation.
