Título: ASSOCIATION BETWEEN SALIVARY PROTEIN ANALYSIS AND ONCOTHERAPY-INDUCED ACUTE ORAL TOXICITY IN PATIENTS WITH ORAL CANCER

Resumo: Objectives: The study tested the hypothesis that targeted proteins are associated with oral toxicities severity in oral and oropharyngeal squamous cell carcinoma (OSCC) patients undergoing head and neck radiotherapy (HNRT).
Study Design: Whole saliva of 17 OSCC participants was collected prior to HNRT. Samples were submitted to liquid biopsy through selected reaction monitoring-mass spectrometry (SRM-MS). Abundance of 56 proteins was correlated with acute oral toxicities (Mann-Whitney and ANOVA with post-hoc Tukey HSD tests): Dysgeusia, dysphagia, odynophagia, oral candidiasis (OC) and xerostomia, which were daily evaluated and classified according to specific scales.
Results: Over half of the participants (76.5%; 64.7% and 64.7%) developed severe dysphagia, dysgeusia and xerostomia, respectively; 47.1% severe odynophagia and 23.5% OC. SRM-MS results indicated that the abundance of 27 proteins, mainly related to biological processes such as innate immune and inflammatory responses, were correlated with more severe grades of dysgeusia, dysphagia, xerostomia, odynophagia, and OC (68%, 60%, 43%, 34%, and 26%, respectively) (p< 0.05).
Conclusions: Present study results are pioneer in characterizing possible salivary biomarkers that may allow the identification of participants that are more likely to develop severe HNRT acute oral toxicities and could help to improve clinical protocols for personalized supportive care in Oncology.
